Description & Mechanism

FOXO4-DRI is a synthetic, modified peptide derived from regions of the Forkhead box O4 (FOXO4) transcription factor. It was engineered using a D-retro-inverso design to improve stability and resistance to proteolytic degradation in experimental systems.

In biochemical and cellular research models, FOXO4-DRI has been used to study protein–protein interactions between FOXO4 and p53. The peptide acts as a competitive inhibitor of this interaction, enabling investigation of transcriptional regulation, cellular senescence pathways, and stress-response signalling. Observed effects are dependent on experimental design, concentration, and model system, and are used for mechanistic exploration rather than defined biological outcomes.

Applications in Research

• Investigation of FOXO4–p53 protein–protein interactions
• Study of cellular senescence and transcription factor regulation
• In vitro models examining stress-response signalling pathways
• Use as a research tool in aging and cell-cycle biology studies

Handling & Stability

• Handle under dry, clean laboratory conditions
• Reconstitute in sterile water or appropriate buffered solution
• Avoid vigorous agitation during dissolution
• Aliquot reconstituted solutions to avoid repeated freeze–thaw cycles
• Store reconstituted material at –20 °C or below for long-term stability
